Omicron SARS-CoV-2 Spike-1 Protein’s Decreased Binding Affinity to ?7nAChr: Implications for Autonomic Dysregulation of the Parasympathetic Nervous System and the Cholinergic Anti-Inflammatory Pathway—An In Silico Analysis

چکیده
Omicron is the dominant strain of COVID-19 in United States and worldwide. Although this variant highly transmissible may evade natural immunity, vaccines, therapeutic antibodies, preclinical results animal models clinical data humans suggest omicron causes a less severe form infection. The molecular basis for attenuation virulence when compared to previous variants currently not well understood. Using protein–ligand docking simulations evaluate compare capacity SARS-CoV-2 spike-1 proteins with different bind human ?7nAChr (i.e., core receptor under control vagus nerve regulating parasympathetic nervous system cholinergic anti-inflammatory pathway), we found that 10 out 14 mutated residues on RBD B.1.1.529 (Omicron) spike, between 0 2 all variants, were present at interaction interface ?7nAChr. We also demonstrated, through protein–protein simulations, these genetic alterations cause dramatic decrease ability protein These suggest, first time, attenuated nature infection animals be attributable particular set alterations, specifically affecting binding site

Evolutionary Analysis of Mammalian ACE2 and the Key Residues Involved in Binding to the Spike Protein Revealed Potential SARS-CoV-2 Hosts
Introduction: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spilled over to humans via wild mammals, entering the host cell using angiotensin-converting enzyme 2 (ACE2) as receptor through Spike (S) protein binding. While SARS-CoV-2 became fully adapted to humans and globally spread, some mammal species were infected back. The present study evaluated the potential risk of mammals...

15 صفحه اولThe effect of temperature on the binding affinity of Remdesivir and RdRp enzyme of SARS-COV-2 virus using steered molecular dynamics simulation
The fatal SARS-COV-2 virus appeared in China at the end of 2019 for the first time. This virus has similar sequence with SARS-COV in 2002, but its infection is very high rate. On the other hand, SARS-COV-2 is a RNA virus and requires RNA-dependent RNA polymerase (RdRp) to transcribe its viral genome. Due to the availability of the active site of this enzyme, an effective treatment is targeting ...
